Pregnanolone Capsules

Pregnanolone (Capsules) – Product Details

Pregnanolone is a naturally occurring steroidal compound that is synthesized in biological systems from cholesterol. It is primarily generated in tissues such as the adrenal glands, brain, and reproductive organs, where it functions as a key intermediate in steroid biosynthesis pathways. Pregnenolone is recognized as a precursor molecule in biochemical research. It is involved in the formation of multiple downstream hormones, including progesterone, cortisol, dehydroepiandrosterone (DHEA), and various androgens and estrogens.

Pregnenolone is also categorized as a neurosteroid because of its presence and activity within neural tissues in experimental conditions. Research shows its involvement in processes such as neuronal signaling, synaptic plasticity, and myelination. Additionally, studies have examined its broader role in steroidogenesis and regulatory pathways linked to hormonal balance.

Note: Pregnenolone is not approved by the FDA for any medical use. Its regulatory status remains unclear and is evaluated on a case-by-case basis, particularly depending on whether it is naturally derived or synthetically produced. It is available for research purposes only and is not intended for clinical or therapeutic use.

Mechanism of Action of Pregnanolone

Pregnanolone is synthesized from cholesterol within mitochondria through enzymatic activity, primarily involving cytochrome P450 enzymes such as CYP11A1. In biochemical research, it is recognized as a key upstream molecule in steroidogenesis. It serves as a substrate for the production of multiple downstream steroid compounds, including progesterone, cortisol, dehydroepiandrosterone (DHEA), and sex steroids. These transformations occur through a series of enzyme-mediated steps that regulate the balance and availability of steroid molecules in experimental systems.

In addition to its role in hormone synthesis, pregnenolone is also classified as a neurosteroid due to its activity within neural tissues. Research indicates that it may interact with neuronal membranes and receptor systems. It influences processes such as synaptic signaling, neurotransmission, and structural support of nerve cells. Experimental findings have also explored its involvement in modulating stress-related pathways through its contribution to cortisol synthesis, as well as its potential role in neuronal protection and plasticity.

Research Findings on Pregnanolone

Anti‑inflammatory and Immune Modulation Effects (Preclinical Research)

In laboratory studies using cultured immune cells, pregnenolone has been shown to influence innate immune signaling pathways at the molecular level. Specifically, pregnenolone was found to promote the targeted degradation of key signaling proteins involved in toll‑like receptor (TLR) pathways, such as TIRAP and TLR2, which play major roles in activating inflammatory responses. This action resulted in reduced secretion of pro‑inflammatory cytokines like tumor necrosis factor‑α (TNFα) and interleukin‑6 (IL‑6). This suggests that pregnenolone can modulate inflammatory signaling in macrophage and microglial cell models studied in vitro.

Neuroactive Steroid Properties in Research Models

In laboratory research, analogs of pregnanolone (a neuroactive form of pregnenolone) have shown effects on nervous system function. For example, studies in developing animal brains demonstrated that pregnanolone glutamate, a derivative compound, can interact with key neurotransmitter receptor systems, such as GABA and NMDA receptors. It also exhibited strong age‑related anticonvulsant effects in seizure models.
