Clomiphene

Product Details – Clomiphene

Clomiphene is a nonsteroidal selective estrogen receptor modulator (SERM) belonging to the triphenylethylene class of compounds. The compound exists as a mixture of geometric isomers (enclomiphene and zuclomiphene), each demonstrating distinct receptor binding characteristics. Clomiphene is frequently applied in in vitro studies to evaluate receptor-ligand interactions and transcriptional regulation mediated by nuclear hormone receptors.

Mechanism of Action

Clomiphene acts as a competitive modulator of estrogen receptors (ERα and ERβ), binding to the ligand-binding domain and altering receptor conformation. This conformational shift influences the recruitment of transcriptional cofactors, resulting in modulation of estrogen-responsive gene expression.

In preclinical models, clomiphene exhibits mixed agonist-antagonist activity depending on cellular context, cofactor availability, and receptor subtype distribution. The presence of its stereoisomers contributes to differential signaling outcomes in research models, making it a valuable compound for studying selective receptor modulation mechanisms.
